She began her acting career in one-act dramas and the sitcom "Nonstop 5" and was also a co-MC with Super Junior's Heechul on SBS's weekly live music show "Inkigayo" from April 23, 2006, to February 18, 2007. Hye Seon soon rose to fame in the drama "Pure in Heart" and surprised critics with her performance in the historical drama "The King and I". Her popularity skyrocketed when she starred in the 2009 series "Boys Over Flowers".

From 2016 to 2019, Gu was married to actor Ahn Jae Hyun, her co-star in the series "Blood". They dated since April 2015 and officially registered their marriage at the Gangnam district office on May 20, 2016, and married on May 21, 2016. They announced that instead of holding a wedding party, they would donate the money to the pediatric ward of Severance Hospital. The pair appeared in the reality show Newlywed Diary which showcased their married life.

On September 1, 2019, she revealed in an Instagram post that she would be taking a break from the entertainment industry to resume college.

She used to be a trainee under SM Entertainment before switching over to DSP Media and preparing to debut in a girl group called Ria, but it didn't work out.

She then signed under YG Entertainment. In June 2019, she signed with HB Entertainment, the same agency as her ex-husband Ahn Jae-hyun. Her contract with HB Entertainment was terminated on April 29, 2020, after it was initiated for her.

On July 23, 2020, she signed with Mimi Entertainment. On May 17, 2021, it was reported that she would be acting, directing, and writing the film “Dark Yellow”, which is being produced in partnership with Wadiz. She has her own production company, Ku Hye Sun Film, which will also produce “Dark Yellow.”

On November 8, 2021, she signed with IOK Company.

On June 18, 2023, it was announced that she had lost the lawsuit she filed against her former agency HB Entertainment for damages in compensation for profits generated from YouTube videos.
